Expression Patterns of Chitinase and Chitosanase, and Degradation Patterns of Chitin-Oligosaccharides by Three Chitinolytic Bacteria

Abstract
In this study, three chitinolytic bacteria were isolated from soil in the rhizosphere of plants: Jeongeupia naejangsanensis GR-1, Paenibacillus gorillae GR-2, and Paenibacillus chitinolyticus GR-3. Among the three strains, P.chitinolyticus GR-3 showed the highest chitinase activity (3.3 U/mL). J. naejangsanensis GR-1 and P. gorillae GR-2 exhibited optimal chitinase activity at pH 5, whereas P. chitinolyticus GR-3 showed optimal activity at pH 4. The three strains had optimal activity at 50°C. The chitinase activity of the three strains decreased in AgNO 3 , SnCl 2 , and CuSO 4 but the activity of P.gorillae GR-2 increased in MgCl 2 and CaCl 2 . Furthermore, the expression patterns of chitinase were confirmed through SDS-PAGE, native-PAGE, and gradient native-PAGE. The degradation patterns of chitinase were confirmed through thin-layer chromatography. J. naejangsanensis GR-1 had exo- and endochitinases, whereas P. gorillae GR-2 and P. chitinolyticus GR-3 had mainly endochitinases. Therefore, we can confirm that the chitinase-producing bacteria J. naejangsanensis GR-1, P. gorillae GR-2, and P. chitinolyticus GR-3 have the potential to be used in agriculture because of their chitin-oligosaccharide production.
